MicroRNA-34a通过靶基因c-Met抑制施万细胞增殖MicroRNA-34a inhibits Schwann cell proliferation by targeting c-Met

摘要(Abstract):
目的:探讨坐骨神经横断后,microRNA-34a(miR-34a)对c-Met(肝细胞生长因子受体)的调控作用及对施万细胞增殖的影响。方法:采用Real-time PCR检测miR-34a和c-Met在坐骨神经横断后近侧端的表达变化;Edu染色法检测miR-34a对原代培养的施万细胞增殖能力的影响;双荧光素酶报告基因系统检测miR-34a对c-Met 3’UTR的直接靶向作用。结果:miR-34a与c-Met在坐骨神经横断后近侧端的表达呈负相关,能够抑制原代培养的施万细胞的增殖,并可直接作用于c-Met基因的3’UTR。结论:miR-34a可以直接靶向c-Met的3’UTR,从而通过下调靶基因c-Met的表达抑制施万细胞的增殖。
